TFPI2 may play a role in the regulation of plasmin-mediated matrix remodeling. It inhibits trypsin, plasmin, factor VIIa/tissue factor and weakly factor Xa, but it has no effect on thrombin. TFPI2 is specific to umbilical vein endothelial cells, liver, placenta, heart, pancreas, and maternal serum at advanced pregnancy. This inhibitor contains three inhibitory domains. TFPI2 contains 3 BPTI/Kunitz inhibitor domains. Function: May play a role in the regulation of plasmin-mediated matrix remodeling. SWISS: P48307 Gene ID: 7980 Database links: Entrez Gene: 7980 Human Omim: 600033 Human SwissProt: P48307 Human Unigene: 438231 Human 組織因子途徑抑制因子(tissue factor pathway in hibitor, TFPI)是組織因子凝血途徑的主要抑制因子， 組織因子途徑抑制因子在諸如動脈粥樣硬化，血管再狹窄，彌漫性血管內(nèi)凝血DIC等方面，有一定的研究價值。 TFPI-2也稱為胎盤蛋白-5(placenta protein 5，PP5)或細胞基質(zhì)聯(lián)合絲氨酸蛋白酶抑制物(matrix associated serine protease inhibitor，MSPI)。 TFPI-2是相對分子質(zhì)量為:25KDa的絲氨酸蛋白酶抑制物, 由脈管系統(tǒng)細胞（內(nèi)皮細胞、平滑肌細胞、成纖維細胞）合成，并沉積在這些細胞的細胞外基質(zhì)里。TFPI-2為廣譜絲氨酸蛋白酶抑制劑，能有效地抑制基質(zhì)金屬蛋白酶（MMP）、纖溶酶、胰蛋白酶、糜蛋白酶、組織蛋白酶等多種蛋白水解酶的活性，在維持細胞外基質(zhì)（ECM）結構完整、抑制腫瘤細胞浸潤轉(zhuǎn)移方面起著重要作用。TFPI-2分布廣泛,在人肝臟、腎臟、心臟、骨骼肌等正常組織中有著高度表達,而在腫瘤組織中也有一定程度的表達。 |
