background: Osteopontin is the principal phosphorylated glycoprotein of bone and is expressed in a limited number of other tissues including dentine. Osteopontin is produced by osteoblasts under stimulation by calcitriol and binds tightly to hydroxyapatite. It is also involved in the anchoring of osteoclasts to the mineral of bone matrix via the vitronectin receptor, which has specificity for osteopontin. Osteopontin is overexpressed in a variety of cancers, including lung, breast, colorectal, stomach, ovarian, melanoma and mesothelioma. Subunit: Ligand for integrin alpha-V/beta-3. Database links: Entrez Gene: 6696 Human Entrez Gene: 20750 Mouse Omim: 166490 Human SwissProt: P10451 Human SwissProt: P10923 Mouse Unigene: 313 Human Important Note: This product as supplied is intended for research use only, not for use in human, therapeutic or diagnostic applications. 骨橋蛋白（osteopontin）又稱為早期Ｔ淋巴細胞活化因子Ｉ或分泌型磷蛋白Ｉ,是一種重要的多功能細胞外基質蛋白,在骨組織細胞、巨噬細胞、平滑肌細胞、內皮細胞及上皮細胞等多種細胞上表達,在介導細胞的趨化、聚集、黏附、增殖和遷移,以及骨組織的礦化與重建、免疫調節(jié)、信號轉導和對感染性疾病的免疫能力等方面有著重要的作用。 ＯＰＮ在細胞免疫中的功能：1.ＯＰＮ對炎性細胞的作用：炎癥是炎性細胞(單核巨噬細胞、淋巴細胞、中性粒細胞等)在炎癥部位的聚集,適度的炎癥對機體有益,如抗感染及促進組織細胞功能恢復等;但過度的炎癥能引起組織損傷等有害作用。ＯＰＮ在炎癥中具有促炎及抗炎雙重作用；2.ＯＰＮ對淋巴細胞的作用：早期的報道稱ＯＰＮ是Ｔ淋巴細胞抑制因子,它可抑制Ｔ淋巴細胞而促進Ｂ淋巴細胞及肥大細胞的分化、增殖 骨橋蛋白(osteopontin，OPN)是一種含精氨酸—甘氨酸—天冬氨酸(Arg-Gly-Asp，RGD)的分泌型糖基化磷蛋白，已歸類于細胞外基質(extracellular matrix，ECM)蛋白。 OPN既是細胞因子，又是細胞黏附分子。 OPN最初是從骨組織分離的，而后在上皮細胞、胃腸道的分泌物、腎、甲狀腺、睪丸以及一些腫瘤細胞里都陸續(xù)的發(fā)現了OPN的表達。在生殖方面，OPN在分泌期子宮內膜上皮細胞、侵襲的滋養(yǎng)細胞、蛻膜細胞中表達。 |
