background: EBNA 3C is a latent viral nuclear protein expressed in EBV transformed lymphoblastic cell lines and is essential for EBV mediated transformation of these cell lines. This viral nuclear protein is essential for EBV mediated positive immunoblastic lymphomas, and functions as a transcription factor. Function: Plays an essential role for the activation and mmortalization of human B-cells. Represses transcription of viral romoters TP1 and Cp through interaction with host RBPJ, and nhibits EBNA2-mediated activation of these promoters. Since Cp is he promoter for all EBNA mRNAs, EBNA6 probably contributes to a egative autoregulatory control loop. Alternatively, EBNA6 also egulates the transcription of the EBV oncogene LMP1 in a cell ycle-dependent manner. It modulates the activity of several host roteins involved in cell cycle regulation including host cyclin A, YC, RB and p27 mainly through binding to the host SCF(SKP2) omplex. Subunit: Interacts with host CTPB1; this interaction seems mportant for EBNA6-mediated transcriptional repression. Interacts ith host MYC; this interaction enhances MYC stability.
