Enhanced green fluorescence protein (EGFP) is a 27 kDa protein derived from the jellyfish Aequorea victoria, which emits green light (emission peak at a wavelenth of 509 nm) when excited by blue light (excitation peak at a wavelenth of 395 nm). EGFP has become an invaluable tool in cell biology research, since its intrinsic fluorescence can be visualized in living cells. EGFP fluorescence is stable under fixation conditions and suitable for a variety of applications. EGFP has been widely used as a reporter for gene expression, enabling researchers to visualize and localize EGFP-tagged proteins within living cells without the need for chemical staining. Other applications of EGFP include assessment of protein protein interactions through the yeast two hybrid system and measurement of distance between proteins through fluorescence energy transfer (FRET) protocols. EGFP technnology has considerably contributed to a greater understanding of cellular physiology. Description: EGFP produced in E.Coli is a single, non-glycosylated, polypeptide chain containing 239 amino acids and having a molecular mass of 27kDa. EGFP is purified by proprietary chromatographic techniques SWISS: P42212 Gene ID: 107331116 綠色熒光蛋白(greenfluorescentprotein,簡稱GFP)是一種能在藍色波長光線激發(fā)下發(fā)出熒光的特殊蛋白質(zhì),正是這種神奇的性質(zhì),讓它成為當(dāng)今生物化學(xué)領(lǐng)域最有力的工具之一,被稱為“生物北斗”。GFP在科學(xué)研究上有著驚人的用途,因為它能夠使我們直接看到細胞內(nèi)部的運動、分布情況。在任何指定的時間我們都可以輕易地找出GFP的定位和存在:你只需要用紫外光去照射,這時所有的GFP都將發(fā)出鮮艷的綠色光芒。顯微鏡下的微觀世界與我們看到的宏觀世界極為不同,而GFP的作用是在微觀與宏觀間架起一座橋梁,讓科學(xué)家通過觀察發(fā)光效應(yīng)推測出物質(zhì)在分子水平上的活動。讓生物化學(xué)家們在研究細胞、蛋白、基因的水平上,從顯微鏡下獲得滿意的結(jié)果。 綠色螢光蛋白在細胞生物學(xué)與分子生物學(xué)領(lǐng)域中,通常被用作為一個報導(dǎo)基因(reporter gene)。一些經(jīng)修飾過的型式可作為生物探針,由于GFP熒光有著生物細胞的自主功能,其熒光的產(chǎn)生不需要任何外源反應(yīng)底物,因此GFP作為一種廣泛應(yīng)用的活體報告蛋白,其作用是任何其它酶類報告的蛋白是無法比擬的。 簡單做個實驗:把GFP連接到你有興趣觀察的任何對象上。比如,你可以把它連接到一種病毒上。然后,隨著病毒在宿主體內(nèi)不斷擴散,你就可以通過跟蹤發(fā)出的綠色螢光來觀察病毒的擴散途徑;或者你把它接合到一種蛋白質(zhì)上并通過顯微鏡觀察它在細胞內(nèi)部的移動或遷移。 目前由我公司自主研發(fā)的重組增強型綠色螢光蛋白及相關(guān)抗體,純度高,穩(wěn)定性好,熒光明亮。 GFP在腫瘤發(fā)病機制研究中的應(yīng)用 GFP是一個分子量較小的蛋白,易與其他一些目的基因形成融合蛋白且不影響自身的目的基因產(chǎn)物的空間構(gòu)象和功能。GFP與目的基因融合,將目的基因標(biāo)記為綠色,若將熒光蛋白植入到動物的某個細胞的DNA分子結(jié)構(gòu)中,可定量分析目的基因的表達水平,顯示其在腫瘤細胞內(nèi)的表達位置和量的變化,為探討該基因在腫瘤發(fā)生、發(fā)展中的作用及其分子機制,提供了便利條件。 在腫瘤的形成過程中,增殖和凋亡是一對相互矛盾的統(tǒng)一體。若腫瘤細胞凋亡占優(yōu)勢,腫瘤組織將長期處于休眠狀態(tài)或自行消亡。腫瘤細胞的凋亡受凋亡相關(guān)基因調(diào)控。用GFP轉(zhuǎn)染腫瘤細胞凋亡相關(guān)基因,并與正常組織進行比較,則大致可判斷此基因為抑制腫瘤細胞凋亡的基因;反之,為促進腫瘤細胞凋亡的基因。 腫瘤細胞浸潤是腫瘤細胞粘連、酶降解、移動和基質(zhì)內(nèi)增殖等一系列過程的表現(xiàn),其根本原因在于腫瘤細胞內(nèi)某些基因表達異常。利用GFP 的示蹤特性,研究腫瘤細胞內(nèi)某些基因異常表達與腫瘤細胞浸潤的關(guān)系,即可揭示腫瘤細胞浸潤的某些機制。 |
